DETECTION OF M.TUBERCULOSIS BY POLYMERASE CHAIN REACTION USING FTC-200

Abstract

|
Polymerase chain reaction (PCR) has been used to detect Mycobacterium tuberculosis for the rapid diagnosis of tuberculosis. In this study, FTC-2000 was used as a thermal cycler and the optimal reaction condition was established with it. A pair of primers which were based on IS6110 repeated sequence known to be specific to M. tuberculosis complex were used. The optimal reaction conditions with FTC-2000 were 100??M of dNTPs each, 0.6??M of primers each, 2.5§¶ of BSA, 0.5 unit of Taq DNA polymerase and 1.0 mM MgCl_2 in 10§¡ reaction mixture. And the denaturation time could be reduced to 1 sec at 94¡É. The 40 reaction cycles were minimally needed for the detectable 123-bps DNA band, but the reaction over 60 cycles was needed for clear DNA band. Under these reaction conditions, the PCR using FTC-2000 may help the rapid detection of the M.tuberculosis.
